  12. 09/03/2011 Pharmacodynamic effects of single oral doses of the novel anxiolytic compound BNC210 in healthy male volunteers

    O'Connor S, Doolin E, Metzger D., Boeijinga P, Pross N, Drutinus E, Andriambeloson E, Kremmidiotis G, Danjou P

BNC210 is a novel agent that exhibits potential utility for the treatment of anxiety and
depression. In preclinical animal models it exhibits anxiolytic and antidepressant effects
without inducing side effects associated with benzodiazepines (BZD) and SSRIs.
A phase I clinical study was conducted at  FORENAP Pharma (France) in healthy male
subjects to investigate the potential side effect profile of BNC210 in comparison to Lorazepam, a BZD commonly used in the treatment of anxiety.

Melatonin is an important regulator of the sleep–wake cycle. A prolonged-release formulation of melatonin (PR-M) that essentially mimics the profile of the endogenous production of the hormone is effective in the treatment of insomnia in patients aged 55 years and older. Because hypnotics result in impairments of various cognitive skills, it is important to examine the cognitive effects associated with the use of PR-M.

  69. 07/11/2008 Are OSAS patients impaired in their driving ability on a circuit with medium traffic density?

    Tassi P, Grenèche J, Pebayle T, Eschenlauer A, Hoeft A, Bonnefond A, Rohmer O, Muzet A.

Many studies have demonstrated that patients with Obstructive Sleep Apnea Syndrome (OSAS), a very common sleep-related breathing disorder, are usually impaired in their driving ability because of decreased sleep quality. However, most of the simulation procedures in laboratories are designed to create monotonic conditions with low traffic density, if any, thereby leading to a dramatic decrease in performance in OSAS patients because of the lack of stimulation. The aim of this study was therefore to evaluate driving abilities in OSAS patients involved in a driving simulation task with medium traffic density, in order to replicate as far as possible real world conditions. The behavioral and physiological attributes likely to predict driving performance in these patients were also investigated. Read more >

  81. 08/22/2007 Insomnia treatment assessment based on physiological data analysis.

    Chouvarda I, Papadelis C, Domis N, Staner L, Maglaveras N.

For the purposes of insomnia treatment, pharmacotherapy is widely used, despite the possibility for the use of behavioural treatment of insomnia. Thus, the assessment and treatment of patients with insomnia needs further investigation. This work addresses insomnia treatment evaluation and medication side-effect assessment based on continuous physiological signals such as EEG and ECG monitoring and analysis. Read more >

  87. 05/10/2007 Ketamine effects on CNS responses assessed with MEG/EEG in a passive auditory sensory-gating paradigm: an attempt for modelling some symptoms of psychosis in man.

Boeijinga PH, Soufflet L, Santoro F, Luthringer R.

Disturbances in integrative function have been consistentLy described in psychotic disorder; for instance, prepulse inhibition of the startle reflex (startle-PPI) which is a marker of sensory gating, is deficient in persons with schizophrenia. The N-methyl-D-aspartate antagonist ketamine produces in control subjects a spectrum of neurobehavioural symptoms like encountered in schizophrenia, and disrupts startle-PPI in animals. Read more >

    Gilles C, Luthringer R.

Animal models of diseases are widely used in the preclinical phase of drug development. They have a place in early human clinical psychopharmacology as well, in order to get early clues that contribute to establish the proof of concept (POC) already in healthy volunteers (HV). Read more >
  89. 04/05/2007 Environmental noise, sleep and health.

    Muzet A.

Unlike other physical ambient factors (i.e. electromagnetic fields or air pollutants), noise is perceived by a specific system (auditory system) in humans. It is therefore a phenomenon that is sensed and evaluated by everybody, and this is why exposure to noise is one of the most, if not the most, frequent complaints of populations living in large cities.

  92. 12/01/2006 Nonlinear regularized wiener filtering with kernel : application in denoising MEG data corrupted by ECG.

    Constantin I, Richard C, Lengellé R, Soufflet L.

Magnetoencephalographic and electroencephalographic recordings are often contaminated by artifacts such as eye movements, blinks, and cardiac or muscle activity. These artifacts, whose amplitude may exceed that of brain signals, may severely interfere with the detection and analysis of events of interest. In this paper, we consider a nonlinear approach for cardiac artifacts removal from magnetoencephalographic data, based on Wiener filtering. Read more >

    Staner L, Boeijinga PH, Danel T, Gendre I, Muzet M, Landron F, Luthringer R.

Sleep disturbances are frequently encountered in alcohol-dependent patients. Drugs improving sleep during abstinence from alcohol may play an important role in the recovery process. In the present study, the effects of acamprosate, a drug successfully used in maintaining abstinence following alcohol withdrawal, were assessed by polysomnographic recordings. Read more >

Recent research suggests that drugs activating nicotine acetylcholine receptors might be promising therapy in cognitive decline seen in the elderly, including Alzheimer's disease. Ispronicline (TC-1734), a brain-selective alpha4beta2 nicotine acetylcholine receptor partial agonist, has shown memory-enhancing properties in rodents and a good tolerability profile. Read more >

    Cerf R.

Macroscopic systems with many interacting subunits, when driven far from equilibrium, exhibit self-organization, for example when a pathological rhythm appears suddenly in an epileptic patient. Sudden changes occurring while conditions vary smoothly have, in cases of interest, underlying mathematics that are the subject of Thom's catastrophe theory. Read more >

  100. 03/01/2006 Surrogate outcomes in neurology, psychiatry, and psychopharmacology.

    Staner L.

A surrogate outcome can be defined as an outcome that can be observed sooner, at lower cost, or less invasively than the true outcome, and that enables valid inferences about the effect of intervention on the true outcome. There is increasing interest in the use of surrogate outcomes of treatment efficacy measurement in investigational drug trials. However, the significance of surrogate markers of treatment outcome in neurology and psychiatry has not yet been sufficiently demonstrated Read more >
